One of the big names who apparently had a major hand in how his character acted was DeForest Kelley. Kelley played Dr. Leonard "Bones" McCoy, the at times dour doctor of the Enterprise.

DeForest Kelley, best remembered as “Star Trek’s” Leonard “Bones” McCoy, died from stomach cancer on June 11, 1999, at the Motion Pictures and Television Fund Hospital in Los Angeles.
